Uncertain significance for Holoprosencephaly 9; Postaxial polydactyly-anterior pituitary anomalies-facial dysmorphism syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001374353.1(GLI2):c.1798G>A (p.Glu600Lys), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with GLI2-related conditions. This sequence change replaces glutamic acid with lysine at codon 617 of the GLI2 protein (p.Glu617Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
