Uncertain significance for Autosomal recessive limb-girdle muscular dystrophy type 2Q; Epidermolysis bullosa simplex, Ogna type; Epidermolysis bullosa simplex with nail dystrophy; Epidermolysis bullosa simplex 5C, with pyloric atresia; Epidermolysis bullosa simplex 5B, with muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_201384.3(PLEC):c.3092C>T (p.Ala1031Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 3092, where C is replaced by T; at the protein level this means replaces alanine at residue 1031 with valine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with PLEC-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces alanine with valine at codon 1058 of the PLEC protein (p.Ala1058Val). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and valine.
