NM_000234.3(LIG1):c.2578G>T (p.Gly860Cys) was classified as Uncertain significance by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with cysteine at codon 860 of the LIG1 protein (p.Gly860Cys). The glycine residue is highly conserved and there is a large physicochemical difference between glycine and cysteine. This variant is present in population databases (rs746525445, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with LIG1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
