Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001366110.1(PAX4):c.467A>G (p.His156Arg), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with PAX4-related conditions. This variant is present in population databases (rs534392927, ExAC 0.01%). This sequence change replaces histidine with arginine at codon 148 of the PAX4 protein (p.His148Arg). The histidine residue is highly conserved and there is a small physicochemical difference between histidine and arginine.
