Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004958.4(MTOR):c.1116+1del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MTOR gene (transcript NM_004958.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1116, deleting one base. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. ClinVar contains an entry for this variant (Variation ID: 1484447). This variant has not been reported in the literature in individuals affected with MTOR-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Val373Cysfs*6) in the MTOR gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in MTOR cause disease.
