Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.2330A>C (p.Gln777Pro), citing Ambry Variant Classification Scheme 2023: The p.Q777P variant (also known as c.2330A>C), located in coding exon 13 of the ALK gene, results from an A to C substitution at nucleotide position 2330. The glutamine at codon 777 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
