Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_016169.4(SUFU):c.1450C>T (p.His484Tyr), citing Ambry Variant Classification Scheme 2023: The p.H484Y variant (also known as c.1450C>T), located in coding exon 12 of the SUFU gene, results from a C to T substitution at nucleotide position 1450. The histidine at codon 484 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
