Uncertain significance for Dyskeratosis congenita — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001099274.3(TINF2):c.144G>C (p.Leu48Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with phenylalanine at codon 48 of the TINF2 protein (p.Leu48Phe). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and phenylalanine. This variant has not been reported in the literature in individuals affected with TINF2-related conditions. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15").
