NM_004329.3(BMPR1A):c.100G>T (p.Gly34Trp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G34W variant (also known as c.100G>T), located in coding exon 2 of the BMPR1A gene, results from a G to T substitution at nucleotide position 100. The glycine at codon 34 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
