Uncertain significance for Nemaline myopathy 10 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_198271.5(LMOD3):c.1130T>C (p.Phe377Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 377 of the LMOD3 protein (p.Phe377Ser). This variant is present in population databases (rs373483102, gnomAD 0.006%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with LMOD3-related conditions. ClinVar contains an entry for this variant (Variation ID: 1483030). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
