Uncertain significance for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma; Arrhythmogenic right ventricular dysplasia 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004415.4(DSP):c.3730A>G (p.Arg1244Gly), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 3730, where A is replaced by G; at the protein level this means replaces arginine at residue 1244 with glycine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals affected with DSP-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with glycine at codon 1244 of the DSP protein (p.Arg1244Gly). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and glycine.
