Submitter rationale: Variant summary: SBF1 c.2643+6C>T alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00021 in 245584 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for a pathogenic variant in SBF1 causing Charcot-Marie-Tooth disease type 4B3 (0.00021 vs 0.0011), allowing no conclusion about variant significance.
