Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002500.5(NEUROD1):c.989T>C (p.Ile330Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 330 of the NEUROD1 protein (p.Ile330Thr). This variant has not been reported in the literature in individuals affected with NEUROD1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (gnomAD no frequency).
