This variant lies in the QARS1 gene (transcript NM_005051.3) at coding-DNA position 793, where C is replaced by T; at the protein level this means replaces arginine at residue 265 with cysteine — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 265 of the QARS protein (p.Arg265Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of microcephaly, progressive, with seizures and cerebral and cerebellar atrophy or early-onset epileptic encephalopathy (PMID: 29875423).
