NM_020975.6(RET):c.53T>C (p.Leu18Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L18P variant (also known as c.53T>C), located in coding exon 1 of the RET gene, results from a T to C substitution at nucleotide position 53. The leucine at codon 18 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
