Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.1873G>A (p.Glu625Lys), citing Ambry Variant Classification Scheme 2023: The p.E625K variant (also known as c.1873G>A), located in coding exon 6 of the AXIN2 gene, results from a G to A substitution at nucleotide position 1873. The glutamic acid at codon 625 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
