Submitter rationale: Variant summary: GJB2 c.585G>T (p.Met195Ile) results in a conservative amino acid change located in the Gap junction protein, cysteine-rich domain (IPR019570) of the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 4e-06 in 251340 control chromosomes. To our knowledge, c.585G>T has not been reported in the literature in individuals affected with autosomal dominant or autosomal recessive nonsyndromic deafness. However, two different variants with the same p.Met195Ile effect (c.585G>C, c.585G>A) have been reported in the heterozygous state in at least 1 individual each with autosomal dominant nonsyndromic deafness (example, Vanniya_2018, Amritkumar_2018, Hochman_2010, Shankar Mani_2009). Further, two different variants affecting the same codon have been classified as likely pathogenic/pathogenic by our lab (p.Met195Val/p.Met195Thr), supporting the critical relevance of codon 195 to GJB2 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
