Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001128840.3(CACNA1D):c.3535G>A (p.Ala1179Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with threonine at codon 1199 of the CACNA1D protein (p.Ala1199Thr). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and threonine. This variant is present in population databases (rs757830356, ExAC 0.01%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with CACNA1D-related conditions.
