Uncertain significance for Hajdu-Cheney syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024408.4(NOTCH2):c.3653G>A (p.Arg1218Gln), citing Invitae Variant Classification Sherloc (09022015): ClinVar contains an entry for this variant (Variation ID: 1479638). This variant has not been reported in the literature in individuals affected with NOTCH2-related conditions. This variant is present in population databases (rs138122751, gnomAD 0.006%). This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 1218 of the NOTCH2 protein (p.Arg1218Gln). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
