NM_004984.4(KIF5A):c.530C>G (p.Pro177Arg) was classified as Uncertain significance for Spastic paraplegia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt KIF5A protein function. This variant has not been reported in the literature in individuals affected with KIF5A-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with arginine at codon 177 of the KIF5A protein (p.Pro177Arg). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and arginine.
