This variant lies in the GSN gene (transcript NM_198252.3) at 1990 bases into the intron immediately before 9 bases upstream of the translation start (5' untranslated region), where G is replaced by T. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with GSN-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with tryptophan at codon 38 of the GSN protein (p.Gly38Trp).
