Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006129.5(BMP1):c.23C>G (p.Pro8Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 8 of the BMP1 protein (p.Pro8Arg). This variant is present in population databases (rs778373466, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with BMP1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1478749). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
