NM_003239.5(TGFB3):c.812A>G (p.Lys271Arg) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K271R variant (also known as c.812A>G), located in coding exon 5 of the TGFB3 gene, results from an A to G substitution at nucleotide position 812. The lysine at codon 271 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
