Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000203.5(IDUA):c.1040G>C (p.Ser347Thr), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the IDUA gene (transcript NM_000203.5) at coding-DNA position 1040, where G is replaced by C; at the protein level this means replaces serine at residue 347 with threonine — a missense variant. Submitter rationale: Variant summary: IDUA c.1040G>C (p.Ser347Thr) results in a conservative amino acid change located in the Glycosyl hydrolases family 39, N-terminal catalytic domain (IPR049166) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 248336 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1040G>C in individuals affected with Mucopolysaccharidosis Type 1 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 1478034). Based on the evidence outlined above, the variant was classified as uncertain significance.
