Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006389.5(HYOU1):c.408T>G (p.Phe136Leu), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs782490652, gnomAD 0.002%). This sequence change replaces phenylalanine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 136 of the HYOU1 protein (p.Phe136Leu). This variant has not been reported in the literature in individuals affected with HYOU1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1477790). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
