Uncertain significance for Nephronophthisis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001128178.3(NPHP1):c.771+158C>T, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with leucine at codon 310 of the NPHP1 protein (p.Pro310Leu). The proline residue is weakly conserved and there is a moderate physicochemical difference between proline and leucine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with NPHP1-related conditions. This variant is not present in population databases (ExAC no frequency).
