This variant lies in the RTEL1 gene (transcript NM_001283009.2) at coding-DNA position 1225, where C is replaced by G; at the protein level this means replaces proline at residue 409 with alanine — a missense variant. Submitter rationale: This sequence change replaces proline, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 409 of the RTEL1 protein (p.Pro409Ala). This variant is present in population databases (rs371603433, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with RTEL1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1477396). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign".
