Uncertain significance for Disseminated atypical mycobacterial infection — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000416.3(IFNGR1):c.569A>C (p.Gln190Pro), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the IFNGR1 gene (transcript NM_000416.3) at coding-DNA position 569, where A is replaced by C; at the protein level this means replaces glutamine at residue 190 with proline — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with IFNGR1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamine with proline at codon 190 of the IFNGR1 protein (p.Gln190Pro). The glutamine residue is weakly conserved and there is a moderate physicochemical difference between glutamine and proline.
