NM_022168.4(IFIH1):c.1159G>A (p.Val387Ile) was classified as Uncertain significance for Aicardi-Goutieres syndrome 7; Singleton-Merten syndrome 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The isoleucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. This variant has not been reported in the literature in individuals affected with IFIH1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces valine with isoleucine at codon 387 of the IFIH1 protein (p.Val387Ile). The valine residue is moderately conserved and there is a small physicochemical difference between valine and isoleucine.
