Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_016169.4(SUFU):c.749A>G (p.His250Arg), citing Ambry Variant Classification Scheme 2023: The p.H250R variant (also known as c.749A>G), located in coding exon 6 of the SUFU gene, results from an A to G substitution at nucleotide position 749. The histidine at codon 250 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
