Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt IQSEC2 protein function. This variant has been observed in individual(s) with clinical features of X-linked intellectual disability (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with glycine at codon 1155 of the IQSEC2 protein (p.Arg1155Gly). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and glycine.
