This variant lies in the POMT1 gene (transcript NM_001077365.2) at coding-DNA position 743, where T is replaced by G; at the protein level this means replaces leucine at residue 248 with arginine — a missense variant. Submitter rationale: This sequence change replaces leucine with arginine at codon 270 of the POMT1 protein (p.Leu270Arg). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and arginine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
