Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_002087.4(GRN):c.620T>C (p.Met207Thr), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the GRN gene (transcript NM_002087.4) at coding-DNA position 620, where T is replaced by C; at the protein level this means replaces methionine at residue 207 with threonine — a missense variant. Submitter rationale: Variant summary: GRN c.620T>C (p.Met207Thr) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2.4e-05 in 251370 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.620T>C has been reported in the literature in individuals affected with Schizophrenia (Geser_2010) and Late-onset Alzheimer Disease (Cruchaga_2012). These reports do not provide unequivocal conclusions about association of the variant with Neuronal Ceroid-Lipofuscinosis (Batten Disease). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
