Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003072.5(SMARCA4):c.4001A>G (p.Lys1334Arg), citing Ambry Variant Classification Scheme 2023: The p.K1334R variant (also known as c.4001A>G), located in coding exon 28 of the SMARCA4 gene, results from an A to G substitution at nucleotide position 4001. The lysine at codon 1334 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
