This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 106969, where G is replaced by A; at the protein level this means replaces glycine at residue 35657 with serine — a missense variant. Submitter rationale: This sequence change replaces glycine with serine at codon 35657 of the TTN protein (p.Gly35657Ser). There is a small physicochemical difference between glycine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with TTN-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
