This variant lies in the KCNJ5 gene (transcript NM_000890.5) at coding-DNA position 224, where G is replaced by A; at the protein level this means replaces serine at residue 75 with asparagine — a missense variant. Submitter rationale: The p.S75N variant (also known as c.224G>A), located in coding exon 1 of the KCNJ5 gene, results from a G to A substitution at nucleotide position 224. The serine at codon 75 is replaced by asparagine, an amino acid with highly similar properties. This variant was detected in a primary aldosteronism cohort, however additional cardiac details were not provided (Gagnon N et al. J Clin Endocrinol Metab, 2018 08;103:2926-2935). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
