This variant is not present in population databases (ExAC no frequency). This sequence change replaces asparagine with isoleucine at codon 153 of the BICD2 protein (p.Asn153Ile). The asparagine residue is moderately conserved and there is a large physicochemical difference between asparagine and isoleucine. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BICD2 protein function.
