NM_001130823.3(DNMT1):c.625A>C (p.Ile209Leu) was classified as Uncertain significance for Hereditary sensory neuropathy-deafness-dementia syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with DNMT1-related conditions. This variant is present in population databases (rs751913403, ExAC 0.001%). This sequence change replaces isoleucine with leucine at codon 209 of the DNMT1 protein (p.Ile209Leu). The isoleucine residue is weakly conserved and there is a small physicochemical difference between isoleucine and leucine.
