Uncertain significance for Beta-D-mannosidosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005908.4(MANBA):c.1610T>A (p.Ile537Asn), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MANBA gene (transcript NM_005908.4) at coding-DNA position 1610, where T is replaced by A; at the protein level this means replaces isoleucine at residue 537 with asparagine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with asparagine at codon 537 of the MANBA protein (p.Ile537Asn). The isoleucine residue is weakly conserved and there is a large physicochemical difference between isoleucine and asparagine. This variant is present in population databases (rs558710124, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with MANBA-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
