Uncertain significance for Muscular dystrophy-dystroglycanopathy (congenital with brain and eye anomalies), type A9; Autosomal recessive limb-girdle muscular dystrophy type 2P — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004393.6(DAG1):c.80C>G (p.Ala27Gly), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DAG1 gene (transcript NM_004393.6) at coding-DNA position 80, where C is replaced by G; at the protein level this means replaces alanine at residue 27 with glycine — a missense variant. Submitter rationale: This sequence change replaces alanine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 27 of the DAG1 protein (p.Ala27Gly). This variant is present in population databases (rs752128921, gnomAD 0.08%). This variant has not been reported in the literature in individuals affected with DAG1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1475256). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
