Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003924.4(PHOX2B):c.866G>A (p.Gly289Asp), citing Ambry Variant Classification Scheme 2023: The p.G289D variant (also known as c.866G>A), located in coding exon 3 of the PHOX2B gene, results from a G to A substitution at nucleotide position 866. The glycine at codon 289 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
