Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003977.4(AIP):c.885C>G (p.Asp295Glu), citing Ambry Variant Classification Scheme 2023: The p.D295E variant (also known as c.885C>G), located in coding exon 6 of the AIP gene, results from a C to G substitution at nucleotide position 885. The aspartic acid at codon 295 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
