Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012193.4(FZD4):c.1552_1555del (p.Lys518fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Lys518Glufs*9) in the FZD4 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 20 amino acid(s) of the FZD4 protein. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with FZD4-related conditions. This variant disrupts a region of the FZD4 protein in which other variant(s) (p.Gly525Arg) have been observed in individuals with FZD4-related conditions (PMID: 20340138). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
