NM_001378454.1(ALMS1):c.173C>G (p.Ser58Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S59C variant (also known as c.176C>G), located in coding exon 1 of the ALMS1 gene, results from a C to G substitution at nucleotide position 176. The serine at codon 59 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
