NM_024334.3(TMEM43):c.706-1G>A was classified as Uncertain Significance for Arrhythmogenic right ventricular dysplasia 5 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This variant causes a G to A nucleotide substitution at the -1 position of intron 8 of the TMEM43 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing. To our knowledge, functional RNA studies have not been reported for this variant. This variant has not been reported in individuals affected with TMEM43-related disorders in the literature. This variant has been identified in 1/249532 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Clinical relevance of loss-of-function truncation and splice variants in the TMEM43 gene is not clearly established. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
