Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.1938G>T (p.Lys646Asn), citing Ambry Variant Classification Scheme 2023: The p.K600N variant (also known as c.1800G>T), located in coding exon 18 of the KIF1B gene, results from a G to T substitution at nucleotide position 1800. The lysine at codon 600 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
