Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.737T>G (p.Val246Gly), citing Ambry Variant Classification Scheme 2023: The p.V246G variant (also known as c.737T>G), located in coding exon 8 of the POLE gene, results from a T to G substitution at nucleotide position 737. The valine at codon 246 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
