Uncertain significance for Autosomal recessive DOPA responsive dystonia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000360.4(TH):c.541A>T (p.Thr181Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with serine at codon 212 of the TH protein (p.Thr212Ser). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and serine. This variant is present in population databases (rs774561953, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with TH-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C55"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
