This variant lies in the GABRA1 gene (transcript NM_001127644.2) at coding-DNA position 326, where C is replaced by T; at the protein level this means replaces threonine at residue 109 with isoleucine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This sequence change replaces threonine with isoleucine at codon 109 of the GABRA1 protein (p.Thr109Ile). The threonine residue is moderately conserved and there is a moderate physicochemical difference between threonine and isoleucine.
