Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001376.5(DYNC1H1):c.12457G>T (p.Val4153Leu), citing Ambry Variant Classification Scheme 2023: The p.V4153L variant (also known as c.12457G>T), located in coding exon 69 of the DYNC1H1 gene, results from a G to T substitution at nucleotide position 12457. The valine at codon 4153 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
